Potent Intradermal Gene Expression of Naked Plasmid DNA in Pig Skin Following Pyro-drive Jet Injection

Intradermal administration of naked DNA with a conventional needle syringe is simple and inexpensive method to expose an encoded antigen the dermal immune system. We aimed enhance intradermal gene expression pyro-drive jet injector using pig skin, which similar in structure biomechanical properties human skin. When Cy3-labeled plasmid (pCy3) was applied skin injector, pCy3 distributed preferentially tissue. Precise localization analysis revealed that also detected intracellular nucleus, frequency substantially higher than syringe. luciferase (pLuc) injected transdermally, activity 380-fold Furthermore, immunohistochemistry showed epidermis positive for protein expression. These data indicate facilitates stable administration, resulting more efficient compared methods. Thus, antigen-expression may provide clinically viable future therapy.
